Bovine pulmonary artery endothelial (BPAE) cells, stained with MitoTracker Red CMXRos, Alexa Fluor 488 phalloidin, and DAPI, were imaged using an image mapping spectrometry (IMS) prototype instrument. The dozen 350 × 350 spectral images represent a total of 48 acquired simultaneously; they show ~0.5 µm resolution and spectral sampling over a 200 nm range with all image detail preserved. Filter sets by Chroma (61001 DAPI/FITC/Propidium Iodide) were used to select the excitation wavelength and separate the fluorescent signal from the excitation light.
