| Powering High-Throughput Next-Generation Imaging The Melles Griot® High Power Laser Engine from IDEX Health & Science is engineered to address the growing need for powerful, cost-effective illumination in fluorescence-based imaging instruments. Why high-power illumination? Companies with high-throughput application platforms in areas like Next Generation Sequencing, Spatial Biology, Next Generation Proteomics, and High Content Screening are striving for even higher throughput. To keep up with the demand for higher throughput, imaging systems are using larger fields of view for step-and-shoot imaging or ultra-fast line scanning with TDI sensors. Both of these methods require significantly higher power illumination to achieve the power densities needed for effective fluorescence detection. While competitors use DPSS technology for these high power systems, they are often too expensive. Our solution provides a cost-effective alternative. The Melles Griot High Power Laser Engine is a compact, cost-effective, multi-wavelength laser diode light engine that delivers high irradiance and photon efficiency. It is specifically designed to enable higher throughput in genomics, transcriptomics, and proteomics workflows. Discover how our modular architecture can seamlessly integrate into your workflow. Market Need for High Power Illumination Higher Throughput Imaging can be achieved in two ways. The first method (Figure 1) applies to the step and shoot imaging using a 2D sensor where a rectangular or square area is illuminated and the fluorescence emission is imaged. The next field of view is then imaged, and this process is repeated until the whole sample is imaged. Then the individual tiles can be stitched together to get data from the whole sample. This process, depending on the application, the sample, the chemistry, and the number of targets to be imaged, can take several hours, or even days, to complete. To increase the throughput, one simply images a larger field of view with a large field of view objective and captures the image with a larger sensor. As the larger field of view is illuminated, the laser power must be increased in order to maintain sufficient irradiance. The second method (Figure 2) uses High speed line scan imaging with a TDI sensor (time delay integration). In this method a Line field of view, typical 10’s of microns in width and a mm in length, is illuminated. The sample stage is moved across at high speed, and the line sensor captures the image in a continuous scan. In this case, since the camera is only seeing the sample for a very short time, more laser power is needed to compensate for shorter exposure times. Current high power illumination solutions in the market use high power DPPS Laser technology (Diode pumped solid state) which are expensive and have a large footprint. Current IDEX Health & Science laser diode solutions only produce power in the range of 500 mW up to 2 W maximum There is, therefore, a market need for a cost-effective high power (> 2 W), multi wavelength, compact Laser engine solution. Optical Efficiency and Etendue Our high power laser engines are engineered to maximize coupling efficiency while minimizing system etendue, ensuring that more of the generated light is delivered to the sample plane—critical for high throughput imaging and detection applications.
